  • How stable is the Clontech In-Fusion enzyme?
    The improved In-Fusion enzyme has been engineered for increased stability. Previously, the enzyme required dilution prior to storage at –20°C, and could only be stored for up to 5 months. The improved In-Fusion enzyme requires no dilution, and can be stored at –20°C (liquid) or room temperature (EcoDry).
  • Why should I use Clontech In-Fusion?
    In-Fusion Cloning Kits allow ligation-independent, directional cloning of PCR products into ANY vector, at ANY site of linearization. These kits provide an easy, rapid, single-tube cloning process.
  • How can I increase transformation efficiency and overall cloning efficiency?
    • Use highly competent E. coli cells that have a transformation efficiency greater than 108 cfu/μg DNA. Most homemade competent cells are not competent enough, especially if the cells are stored before use. We recommend  Stellar™ Competent Cells (Cat. No. 636763) which are optimized for use with In-Fusion.
    • It is important that the PCR fragment be purified away from dNTPs after PCR.
    • If you are not using a commercial purification kit, gel purification should be followed by phenol chloroform extraction to remove any impurities.
    • The primers must be of good quality to ensure the sequence of the homologous region is correct.
